Primers amplifying unique bands (from Step 3) are tested for polymorphisms between Nigerian and Ivory Coast Strains.

The P1 individuals (NF7 male and ICF5 female) are included in this analysis to determine which markers will be useful for the mapcross analysis.

Sample polymorphism gel. ³²P-dCTP was added to PCR reactions and samples were electrophoresed on 6% acrylamide sequencing gels to separate alleles. Included are 2 unrelated Nigerian (N) and 2 unrelated Ivory Coast (IC) individuals. Np and ICp represent the P1s used in the mapcross (Step 2).
